My first couple of times ground collecting an older gentle stallion and the semen wasnt white but opaque / first time very little w/ lots of gel/ second time increased pressure and temp to 135 h2o going into the lumen/bladder. then on collection lots of opaque "semen fraction" ( half the baby bottle liner) and not so much gel/ check under scope and there were sperm in it/ didnt smell like urine/ what am I doing wrong in AV setup as this gentleman was collected and frozen w/ good results.??? When exactly do you slip the AV on the stallion? When you see the preejaculate clear fluid?? IF the AV is correct shouldnt the stallion "fire right off"? Wish I could talk horse so he could tell me what I am doing wrong...

I'm not quite sure wat you're asking here... :-)

If you're getting an ejaculate containing sperm, then you must be doing it right!

You would put the AV on once you see that the stallion has an erection and appears willing to breed - you don't have to wait for the appearance of pre-ejaculatory fluid.

Some stallions will get right down to work, and others will require repeated attempts - there's no universal rule.

135° might be a little cooler than he likes - the normal range for final equilibrated internal temperature is 113-130°, so if you're putting it in at 135° you have to remember that it's going to cool down and probably be only about 120° or so.

If I'm missing the point, please don't hesitate to clarify what it is that you're asking!!! :-)

Well, I thought that the semen fraction was supposed to be white not an opaque vanilla color. that I guess is my main concern as I have seen the end result of others and they were white not a pale pale yellow and skim milk like.Is there anything I can do to improve the quality?? or does it really matter if they are viably good sperm? Just breed and dont worry about it? I dont have the hemocytometer yet/should tomorrow for an actual count. The sperm were definitely forward moving under the scope and no messed up tails or circler's. I thought perhaps I had something very wrong as it wasnt coming out white semen fraction/ the gel is white and egg white like stuff though. Per classes I am/was concerned that it wasnt where it should be because it wasnt white .... any comments?

The non-magnified appearance of semen will vary somewhat depending upon the concentration. The more highly concentrated it is, the more opaque and often slightly "creamier" in colour. The less concentrated, the more it looks like skim milk - almost a bluish tint to it, and a degree of transparency. In other words, all ejaculates are not created equal... :-)

The gel fraction will typically appear like the egg white you mention, and comes as the end portion of the ejaculate - a useful indicator that you have got a complete ejaculation is the presence of gel fraction (note that not all stallions produce gel, so the absence doesn't necessarily mean that you didn't get a complete ejaculate!).

In all probability if there are motile sperm there, you're just fine. Get that hemacytometer working and you'll probably start to equate what the raw semen looks like with the concentration... :-)

Hemocytometer... I need help with the formulas for the counts. Can anyone help me with this? I want to have it wrote down to be able to follow it. I finally got how to use the silly thing, but need to get all the counts. Thanks!

We actually go over this in detail at our courses, and it is quite simple when you get the hang of it!

There are a couple of links in the link section of the web site to other sites that describe the use of the hemacytometer to count sperm, but in essence, dilute at 1:99, count 25 squares (13 on one grid and 12 on the other) and the number of sperm you count will be the concentration in millions/ml. If you count all 50 squares, then divide by 2 and the count will be in millions/ml.
