My computer is not in the lab. Do you have a formula for computing extender that I can put on the wall and use when needed???

  • You need to send a minimum of one billion sperm, presuming normal morphology and progressive motility at the time of collection.
  • You need a final concentration of between 25 and 50 million sperm/ml once extended.
  • You need a minimum extender:semen ratio of 3:1 if using a Kenney-based extender.
  • You will need a calculator.
  • The above only applies if the semen is "normal".

When getting ready for shipped use, would you like to me drawl up the semen and then the bathed extender or do you want me to mix together and then drawl???

The faster that you dilute the toxic effects of seminal plasma, the better off that you are. What we will typically do is immediately after collection we add an equal volume of extender to the filtered ejaculate, after having removed a small sample for evaluation (sperm count, motility etc.). This then allows us some time to complete the evaluations and calculations to determine how much final volume of semen we need to send and how much more extender we need to add.

Can you please tell me the best way to package semen for the Equitainer? Being it has been extended and ready for shipment.

Once extended, the semen can be packaged in either 50-ml centrifuge tubes or double-bagged in Whirl-Pak bags. You can also use baby bottle liners if you can find them (Playtex has stopped making the older-style bags, but there are some non-name-brand ones available). If using the bags, you can close them with a heat sealer or double rubber (elastic) bands.

Can you tell me please the best way to count motility using the microscope???

Do you know anyone that sells an AI kit? Have seen one for canine.

Dilute the raw semen until you are looking at perhaps only 20 sperm under each microscope view, then do an estimate of how many sperm are moving, then how many are moving in a straight line, then calculate the % motility and progressive motility respectively.

I believe Exodus Breeders Supply carries them.

We had a clients veterinarian request a 50ml dose with 50million sperm instead of 25. We used your calculator and are confused with the warning. Could you please walk us through a calculation to see if we are correct in our collection/extension. The collection yield a concentration of 488 mil sperm with 65% progressive motility. To ship 50 ml with 50 million sperm we calculated 6.3 ml raw semen and 43.7 extender. We just want to get this project done for the stallion owner. Please Help!

I'm not completely clear exactly what it is you're trying to get...

If you want to calculate a final concentration of 50 million sperm/ml then all you need to do is change the entry at "Final Desired Extended Concentration (in millions of sperm per ml):" to "50" from the default value of "25". If you do that, based upon an initial concentration of 488 x 106/ml with 65% PM, it returns a requirement of 2 ml raw semen diluted 9.8 times (it's slightly more than 2 ml raw semen, so you would probably round that up to 3 ml) - this equates to 1 part of semen to 8.8 parts of extender (2.0 mls raw semen and 18.0 mls of extender or more likely round it up to 3 ml raw semen to 26 ml extender).

If you are shipping 1 billion sperm (a standard insemination dose), it is not possible to ship a 50-ml dose at 50 million/ml (50 ml at 50 m/ml = 2.5 billion) which is why the calculator will not return a 50-ml total volume calculation. You will either have to ship a lower volume (as above - 20 or 29 ml depending which you choose), or a higher number of sperm.

If you decide to go for the higher number of sperm, then you can ship 2.5 billion, which will require about 5 ml raw semen (actually 2.44 billion at your given raw concentration) and 45 ml extender. This would actually yield a final concentration of 48.8 million sperm/ml.

OK JOS, I'm confussed, will there be enough for the sperm to eat while being shipped if you double the concentration of sperm? Are they doubling up on the concentration because the survival rate is poor? Do they make a more concentrated extender that has more "sperm food" per ml. for sending a more sperm concentrated dose?

The desired final concentration for shipping once diluted is between 25 and 50 million sperm per ml. It is therefore to be hoped that increasing the concentration with this stallion will not have a deleterious effect. With some stallions it will make no difference, with others it can.

As far as "why?" - you'll have to ask them that! :-)
