Semen Sexing

Postby friesiangal » Tue Dec 10, 2013 3:47 pm

Hello
We were recently discussing sexing the semen. Is there a different G force and timeing that would be needed to spin to separate the male to female semen and which layers would be selected for each. The two rates I'm familiar with is 400g/12 min for non cushioned and 1000g/20 min for cushioned. Are there any rate studies on accuracy for female vs male born to know if it's worth attempting the selection pre insemination.

The only reliable method for sexing semen is use of a process incorporating a flow cytometer. Flow cytometers cost upwards of $1,000,000! (And even that technique is only 90% successful!).

There are people that will tell you that centrifuging semen "sexes" it, but there is no peer-reviewed research that shows that it does.

In keeping with the centrifuge conversation, to concentrate good semen or pull out the dead stuff what is the best g force or speed/time to spin, would it need cushion or be ok without? I know for freezing it's 400g for non cushion and 1000g with cushion but for "cleaning out" the junk for chilled shipping what would be recommended

In order to effectively remove dead or damaged sperm, one needs to use a gradient solution such as Androcol-E. This is typically centrifuged at 400G. It is important to note that it will not remove dead sperm that are present as a result of spermiostasis as they will spin down into the live sperm layer as well.
